Effect of fetal bovine serum on the growth of hemocytes culture of Helicoverpa armigera, Ephestia kuehniella and Arge ochropus

چکیده
Since the use of insect cell lines for producing bio-pesticides and recombinant protein expression or gene delivery is increasing, the establishment of new cell lines will help to enrich this industry. Therefore, several primary cultures were initiated from hemocytes of Helicoverpa armigera (Hubner), Ephestia kuehniella Zeller and Arge ochropus (Gmelin). The cultures were incubated in EX-cell 400 medium supplemented with 0, 5, 10, 15 and 20% concentrations of fetal bovine serum (FBS) at 28 ºC .These concentrations of FBS were chosen arbitrarily by us as well as by others working on insect tissue culture. Cell viability was low and no growth occurred in any of the cell lines when FBS was omitted from the medium. Maximum growth of H. armigera hemocytes was obtained with 15% FBS. There was no further increase in this growth when FBS was increased to 20%, and neither did the increased concentrations have any appreciable adverse effect on the growth or survival of these cells. Maximum growth of E. kuehniella hemocytes was obtained in 10% FBS; the growth of these cells was significantly less in 15 and 20% FBS. Growth of A. ochropus hemocytes was rich when FBS concentration was increased to these levels. In conclusion, the current investigation demonstrated the lowest concentration of FBS required for maximum growth of insect cells in vitro
