Report of accumulation aldose reductase from gut of recurrent sunn pest, Eurygaster integriceps using proteomic technique

The sorbitol pathway includes converts process of glucose to sorbitol and then fructose. This metabolic pathway reported from human and insects. The key enzyme in sorbitol forming is aldose reductase (EC 1.1.1.21). One-day-old adult insects were selected for dissection of digestive system. The guts were transferred to microtubes containing phosphate-buffered saline (pH: 6.9) and a cocktail of protease Inhibitors. Proteins were extracted by the acetone/trichloroacetic acid method. A total of 300 μg proteins of the guts were applied to immobilized pH (IPG) strips (11 cm, pH 3-10 linear) to perform two dimensional electrophoresis (2DE). One of the main spots that not identified but exist in all of replication was selected as target protein. This spot cut and digest with trypsin and then injected to mass spectrometry (LC-MS/MS). After analysis of resulted spectrums using National Center of Biotechnology Information database with two entries Animals and Wheat, target protein identifies as aldose reductase with accession number of gi 115615333. Aldose reductase is an oxidoreductase enzyme that converts glucose to sorbitol as the first reaction in the polyol pathway in the animal world. Our result proves that effective proteins in the sorbitol pathway accumulated in the gut of recurrent sunn pest. Sorbitol as critical carbohydrate is necessary for survival of overwintering adults. On the other hand, aldose reductase as a main enzyme in the carbohydrate metabolism has a key role in the biochemical process free ATP. Hence, any disruption of the sorbitol forming may be causes increase mortality of sunn pest in the overwintering stage. Next step in the gut physiology studies of the adult sunn pest will be focusing on the improvement and synthesis of aldose reductase inhibitors to disrupt sorbitol forming in the gut of sunn pest. On the other hand, it is needed to our knowledge increased about gut proteins and their effects on the overwintering stage in the sunn pest and the similar insects with identification of new keys proteins in the various metabolisms pathway with proteomics and the other related techniques.
