Genetic diversity of some Japanese races of silkworm Bombyx mori by ISSR ‎‎(inter-simple sequence repeat) molecular marker

Identification of biodiversity in the silkworm is also important for designing better Breeding ‎programs Genetic diversity of five Japanese races of silkworm Bombyx mori stored in Iran Silkworm ‎germplasm were studied by molecular marker ISSR. Silkworm races reared in Iran Silkworm ‎Research Center (Rasht, Iran) according standard methods. Six adults collected for DNA extraction ‎in order to study genetic diversity. PCR products loaded on 1.5% agarose gel for 3 h in 80v and ‎stained by ethidium bromide for visualizing by gel documentation. Number of monomorphic and ‎polymorphic produced fragments, polymorphism percentage, polymorphic information content, ‎marker index, resolving power, effective multiplex ratio were calculated. Cluster analysis of five ‎population obtained by NTSYS 2.01 software. Results showed twelve used primers produced 156 ‎scorable bands with fragment size between 250-3000 bp. Obtained dendrogram inserted studied ‎populations in three various groups accordance with geographical origin. It seems that ISSR ‎molecular marker could discriminated geographical races of Japanese lines.‎
